Born in [[Lallio]],<ref name=figc>{{cite web|url=http://www.figc.it/nazionali/DettaglioConvocato?codiceConvocato=406&squadra=1|title=Convocazioni e presenze in campo|publisher=FIGC|language=it|access-date=2 January 2015|archive-url=https://web.archive.org/web/20150427143045/http://www.figc.it/nazionali/DettaglioConvocato?codiceConvocato=406&squadra=1|archive-date=27 April 2015|url-status=dead}}</ref> [[province of Bergamo]], he began his professional career with his local club [[Atalanta B.C.|Atalanta]] in 1960, winning the [[Coppa Italia]] in 1963, and finishing the tournament as the top goalscorer with five goals, also scoring a hat-trick in the final against [[Torino F.C.|Torino]].<ref name="Storie di Calcio: Domenghini"/> He then moved to [[Inter Milan]] in 1964, and was a member of manager [[Helenio Herrera]]'s highly successful "[[Grande Inter]]" squad; he played 164 times for the club, scoring 54 goals, winning two [[Serie A]] titles, two [[Intercontinental Cup (1960–2004)|Intercontinental Cups]], and a [[European Cup]].

He also later played for [[Cagliari Calcio|Cagliari]], forming a notable attacking trio alongside [[Luigi Riva|Gigi Riva]] and [[Sergio Gori|Sergio "Bobo" Gori]], as they went on to win the Serie A title during the [[1969–70 Serie A|1969–70 season]], Domenghini's first season with the club. He later played with [[A.S. Roma]] for the [[1973–74 Serie A|1973–74 season]], subsequently moving [[Serie B]] side [[Hellas Verona F.C.|Verona]] in [[1974–75 Serie B|1974]], for two seasons, helping them gain promotion to [[1975–76 Serie A|Serie A in 1975]]. He then played one more [[1976–77 Serie A|final season]] in Serie A with [[Foggia Calcio|Foggia]], before playing in the [[Serie C]] lower divisions, with [[Olbia Calcio|Olbia]] for [[1977–78 Serie C|one season]], and subsequently for [[Trento Calcio 1921|Trento]] for a single [[Serie C1]] season, before retiring in 1979.
